Summary of Position
The intern will be developing a new, highly efficient molecular technique to create mutations in plasmids for the purpose of antibody engineering. Current methods to construct large synthetic antibody libraries are hindered by low transformation efficiency (PCR-digestion-ligation method), low recombination rate (Kunkel method) or a multi-step process with limited application (Pfunkel and Kunkel+RCA), thus a new method is needed that can easily insert mutations with high recombination rate. The method should be scalable to generate large quantities of mutagenic DNA with high transformation efficiency for creating a large synthetic antibody library.
